Jesse Spencer est né le 12 février 1979 à Melbourne en Australie. Jesse a deux frères aînés ( Tarney et Luc) et une sœur cadette (Polly). Il joue de la guitare, du piano et également du violon depuis ses 10 ans. Ses frères et son père sont tous docteurs.

A l'âge de 8 ans, il a rejoint une choral

En 1998, il est la voix de Triton Jr dans le dessin animé de Disney « Hercules ». Il enchaîne ensuite plusieurs rôles à la télévision et quelques apparitions au cinéma avant d'intégrer le casting de « House M.D. » sous les traits du Dr Robert Chase.

Jesse fut nominé en 1999 pour le " 1999 Silver Logie " dans la catégorie de l'acteur le plus populaire.
